    Two BD FACS Aria II Cell Sorters/Analyzers

    • Dedicated operator required 
    • A four laser Aria II (UV/407nm, 488nm, 532nm, 594nm) for Research Flow Cytometry is capable of detecting fifteen colors (FL, Cy55PerCP, PE, TRPE, Cy5PE, Cy55PE, Cy7PE, TR, APC, Cy55APC, Cy7APC, CasBlue, BV 510, BV 785, QDot605) + two scatter channels. 
    • Another four laser Aria II is housed in a biosafety hood and fitted with appropriate filtration equipment to protect personnel during analysis of human samples for the Human Immune Monitoring services. Laser emission spectra are 355 nm, 405 nm, 488 nm, and 633 nm with capability of detecting 14 colors (FITC, PE, APC, PerCP/Cy5.5PE, Alexafluor 700, Pacific Blue, Cy7/PE, BUV395, BUV737, Cy7/APC, PerCP, BV605,  BV510, and BUV496) + two scatter channels.

    Miltenyi gentleMACS Tissue Dissociator

    The gentleMACS Dissociator dissociates tissues, such as tumor samples, into single cell suspensions for subsequent analysis or sorting of individual cell subsets by flow cytometry. The equipment uses optimized protocols to dissociate and homogenize up to two tissues samples in a semi-automated manner, resulting in reproducible tissue disruption and single cell release.

    Isoplexis IsoSpark

    • This instrument can isolate individual cells and perform single-cell proteomic analysis 
    • Kits from Isoplexus can detect secretion of cytokines and chemokines or intracellular expression of phosphoproteins or transcription factors 

    BD Symphony S6 Spectral Sorter/Analyzer

    • Dedicated operator required.
    • Five laser (355nm, 405nm, 488nm, 561nm, 637nm) twenty-eight color analyzer with optional HTS resource.
    • Spectral detection with 48 PMTs
    • Sorts up to six distinct streams of cells.
    • Housed in a biosafety hood and fitted with appropriate filtration equipment to protect personnel during analysis of human samples for the Human Immune Monitoring services

    Miltenyi MultiMACS Cell24 Magnetic Separation Device

    The MACS unit can be used for enriching or depleting cell populations prior to final sorting by flow cytometry.  Mixed cell populations, such as splenocytes or peripheral blood mononuclear cells, can be exposed to superparamagnetic nano-sized beads conjugated to monoclonal antibodies targeting surface biomarkers on specific cell subsets. Then the labeled cells are passed over a high magnetic gradient in MACS columns to achieve positive cell selection or negative cell depletion from the starting cell mixture.

    Computers

    The laboratory has two desktop Dell Precision Tower 5810 PC computer workstations and a Hewlett Packard LaserJet Enterprise M553 color printer that are connected by ethernet to internet services and and the data storage Flow-LIMS system.  Software includes Microsoft Office, FlowJo software for flow cytometry data analysis, Mat Lab for statistical analysis, Prism for data analysis and plotting, and Corel Draw for figure preparation.
